Beta-amyloid
A small peptide fragment produced by enzymatic cleavage of amyloid precursor protein. In the brains of patients with Alzheimer's disease it aggregates abnormally to form oligomers and plaques, and it is considered neurotoxic.
Timeline (last 90 days)
The glymphatic system differs from the lymphatic system: it does not transport immune cells, but instead uses the exchange between cerebrospinal fluid and brain interstitial fluid to drain metabolic products such as beta-amyloid and tau protein out of the brain.
A new study claims that restoring just two hours of sleep can improve certain functional performance without clearing brain amyloid plaques.
The clearance of beta-amyloid depends heavily on glymphatic system activity during deep sleep.
Beta-amyloid is a small peptide fragment produced by enzymatic cleavage of amyloid precursor protein (APP); under normal physiological conditions it can be cleared, but in the brains of Alzheimer's disease patients it abnormally aggregates to form oligomers and plaques.
The mechanism of action of lecanemab and donanemab is to target and clear abnormally accumulated β-amyloid plaques in the brain, and both belong to monoclonal antibody therapies
The glymphatic system is a brain waste-clearance mechanism discovered by the scientific community in 2013; it is primarily active during sleep and removes metabolic waste products such as β-amyloid and tau proteins through cerebrospinal fluid flow
During deep non-rapid eye movement (NREM) sleep, the brain's glymphatic system clears metabolic waste products, including β-amyloid associated with Alzheimer's disease.
A positive amyloid pathology biomarker does not mean an individual will necessarily develop Alzheimer's disease dementia, and there is a risk of overdiagnosis.
